Toby,
You break the action open (similar to a Harrison and Richardson ). You then place a cartridge in the hole that's at the end of the barrel closest to the pistol grip, i.e. breach. Next you close the action. Point the firearm in a safe direction or an intended target. Cock the hammer (that thing in top of the metal above the trigger guard) then squeeze the trigger SLOWLY and it should go boom.

What happens if you load it from the muzzle and fire it at an unintended target?

For what it's worth I know of 1 person with a Henry single shot, I don't remember the caliber, but he was pretty fond of it. I remember he put a vxii on top though.
